| 概要 | This review traces melatonin research in Alzheimer’s disease from preclinical mechanisms to emerging therapies. It summarizes evidence that melatonin may reduce oxidative stress, neuroinflammation, amyloid and tau pathology, while supporting sleep, circadian regulation, and cognition. Clinical studies have examined sleep, behavior, cognitive outcomes, electrophysiology, and biomarkers in mild cognitive impairment and Alzheimer’s disease, but remain heterogeneous and limited. The article also evaluates melatonin analogues, derivatives, and hybrid molecules designed to improve potency or target multiple pathways. Overall, the therapeutic rationale is strong, but definitive clinical trials are still needed. |
