| 概要 | This review assesses the orexin system as a mechanistic and therapeutic target in depression. Orexin neurons influence sleep–wake regulation, reward, motivation, stress responses, and hypothalamic–pituitary–adrenal activity, all of which can be disrupted in depression. The article distinguishes functional roles of dorsomedial and perifornical orexin populations from lateral hypothalamic populations and considers receptor-specific effects. Although altered orexin signaling may help explain particular symptoms, findings across models are sometimes conflicting. The review supports targeted, symptom-sensitive investigation rather than assuming that global orexin activation or blockade will be uniformly beneficial. |
